Given the complexity of metabolic networks, identification of optimal metabolic intervention strategies for redirecting fluxes towards desired products is a challenging task. Several algorithms based on linear programming and pathway analysis have been proposed. However, there is still a lack of an algorithmic framework that exploits the range of optimal and suboptimal routes and the structural/regulatory properties thereof. To this end, we are using a modified version of the concept “control effective flux (CEF)” [1] towards a novel algorithm for in silico metabolic engineering. CEFs represent the importance of each reaction for efficient and flexible operation of the entire metabolic network.

Objective: Trichophyton rubrum is presently the most common
worldwide pathogen causing dermatophytoses such as tinea corporis,
tinea capitis, tinea pedis, and onychomycosis [1]. The main aim
of the present work was assess MALDI-TOF ICMS as a fast and
reliable technique in the identification of T. rubrum from clinical
cases occurrence in the Portuguese health centres, and evaluates the
potentialities and limits of this new microbial identification technique
on the taxonomy of these infectious dermatophytes.
Methods: Fungi were grown for 10 days in solid medium (SDA,
Sabouraud Dextrose Agar) and then the mycelia were direct transferred
from the SDA plate to the MALDI stainless steel template and mixed
with 1 ml MALDI matrix solution (75 mg/ml 2,5-dihydroxybenzoic acid
in ethanol/water/acetonitrile [1:1:1] with 0.03% trifluoroacetic acid)...
Biofilms are structurally complex microconsortia of surface adhering cells embedded within
an extracellular matrix (ECM) composed of substances produced and secreted by cells or
derived from cell lysis. One of the recently discovered bacterial biofilms ECM components is
the extracellular DNA (eDNA). Although the investigation on eDNA in fungal biofilms is
scarce, preliminary studies suggest that eDNA may play a role in biofilms formed by the
opportunistic fungal pathogen Candida albicans. Thus, the present study aimed at
determining the eDNA content of C. albicans SC5314 biofilm ECM and the effect of DNase I
treatment on biofilm formation and biofilm cells susceptibility to antifungals, as indicators of
the role of eDNA in C. albicans biofilms.

Olive mill wastewater (OMW) is a liquid waste that results in large amounts from the olive oil manufacturing industry. The quality and quantity of OMW constituents are dependent of many factors, such as olives type and maturity, climatic conditions and region of origin, cultivation methods, and technology used for oil extraction1. From the 3-phases centrifugation process around 1.6 cubic meters of OMW per ton of olives processed are generated. São apresentados os fundamentos dos métodos de cálculo de chumaceiras radiais hidrodinâmicas. As características de desempenho da chumaceira obtidas por utilização de três dos métodos mais vulgarmente utilizados na prática, e representativos de categorias distintas, são comparadas entre si. Esta análise comparativa é feita com base nas previsões teóricas de parâmetros de desempenho (nomeadamente, caudal de óleo na chumceira, potência dissipada por atrito viscoso e temperatura máxima) e em resultados experimentais publicados. Na presente comunicação é apresentada uma análise teórica do comportamento termohidrodinâmico de chumaceiras radiais dotadas de um sulco de alimentação radial. O modelo numérico desenvolvido para o efeito baseia-se no algoritmo de Elrod e permite obter uma solução para a distribuição de pressões hidrodinâmicas na chumaceira. Esta solução é obtida em simultaneidade com a distribuição de temperaturas no fluido, no veio e no casquilho. O procedimento computacional permite simular convenientemente a cavitação, a regeneração e a transferência de calor na chumaceira. As previsões teóricas obtidas são comparadas com resultados experimentais disponíveis na literatura. É discutida a influência de alguns parâmetros de alimentação nas características de desempenho da chumaceira, nomeadamente no caudal e na temperatura máxima.
